Admittedly I don't have much room to compare (besides a very short lived MA/Regen) but I love my DM/EA/Lev stalker. Hits like a speeding semi and never stops.
Love my Elec/Nin/Mu. AoE of death. So much fun to play.
DM/EA that runs a close to 4 bil inf build. No staminia and has enough end to solo AVs, and then some
pvpo'd spines/regin with no stamina and leadership.
even though i understand that in some situations, it is definitely suboptimal.

I only have one stalker, NRG/NRG - no epics. Was fun in pvp early on, since all the changes, not so much, but in pve, other than AV's I'm pretty much untouchable.
I think overall, I would like a spines/regen. What I currently "have" is a 50 em/regen and a 50 spines/nin. I prefer the spines/nin and no longer enjoy my em/regen. Still considering deleting the em/regen and re-rolling to the spines/regen.
PvE - Elec/WP, with tough and + end bonus'...feels like a Scrapper. No need to hit, run, hide and repeat...I know its blasphemy, but I enjoy duking it out.

personaly my main toon is a clw/nin and i love it, it is not good for min/max-er though. clws have great rech and the range powers realy make up for the lower dps.
I like my spines/wp.

My Elec/Elec softcapped to melee is my favorite to play.. I can roll through anything in the game whatsoever. Working on a new BS/SR that may be second place when I'm done.

My Dark/Dark/Soul Stalker is nice, he can withstand Romulus' rez, Shadow Cysts exploding, 2 of the best self heals in the game, pure awsome.
DM/SR is tastyness, and very much the Assassin.
Claws/Will/Soul is a fury-less brute. Sooo much fun.
The rest of mine are too low to tell as of now.

Well Ive been away for 8 months or so but I fell straight back into my EM / Nin just like I never left.
On a side note not much seems to have changed since I left other than the very obvious lack of people playing the game. : (

I like MA/DA its got awesome sauce all over it.
The only stalker I have not gotten bored with... NB/EA.
He is only lvl 22 so far, but I heart it.

It definitely plays like the sword-wielding ninja assassin stereotype
we all know from the movies... 
It's still the perfect blend of effectiveness and RP concept for me.
