Most damage ever done with one shot?
Not mine, but during an eden trial with some friends, the fireblaster manage to dinged off about 2400 points of damage on a single inferno.

Back when it first came out as I recall, the Lady Grey Negative Energy proc was bugged and doing huge amounts of damage. Many thousands IIRC.

i believe the winner would be that peacebringer a long time ago, that was able to put his slots in 2 attacks when respec'd. He 2 shotted Hami and got the Bug Hunter Badge.

If I remember correctly the most damage ever done in 1 shot is when one of the houses in Steel Canyon actually burns down.
Do multiple Trip Mines count?
Cause if they do I was able to take out a Ballista in one shot.
Many Millions, actually. I think Local Man has a screenie of the highest single damage point ever recorded. It was so high the game had to use scientific notation to display it correctly.

I think the highest non-glitch damage has to go to Inferno. When slotted and with Aim and Build Up, that puppy can burn a heckuva lot of hp.

Okay, maybe not scientific notation big, but remember the seas of orange numbers floating up from the old dumpster diving days before the aggro limits and AoE target limits?

I'll bet a damage-capped Inferno would do some pretty intense damage to a level 1 Hellion with his resist floored.
Maybe not as much as the "glitched" damages, but probably the highest legit damage, if you're counting one-shots not over time/multiple hits like Blizzard would be.

I'm pretty sure, excluding bugs, the best case scenario is a damage-capped corruptor with a PBAoE nuke against a res-floored level 1 that is already injured.
That ought to be roughly:
~41.71 (corruptor base damage at 50) x 6 (DS for all three nuke hits) x 5 (buff cap) x 6 (level difference) x 4 (floored res) x 2 (scourge) = 60062.4
Inferno's extra DoT would push it a lot higher if it all hit, but of course nothing would survive to see it.
I don't know, cause if your going to be at damage cap,have the enemies resist in the negatives and lvl 50 vs lvl 1 I think the Double Assassins could probably match that.
Of course the Stalker can't set that up them self, but that's not relevant to the discussion is it?
I ask because I just did this:
