Resistance vs Defense
Not much, depending on what side you're on. Villain-side has a bit more than hero-side, but if you know which mobs to look out for, you can handle it.
Also, Resistance resists resistance debuffs, so it's got that going for it. Still, I prefer Defense over Resistance, since if you block the attack completely, you avoid any secondary effects, too.

Resistance is hard to obtain from set bonuses and pool powers. Defense is easy to obtain from set bonuses and pool powers. Stacking defense on a resistance-based character makes it much more durable than going for only one type of mitigation.
Another type of mitigation is healing. Willpower has a lot of regeneration. Stone Armor has a decent amount of it with Rooted on.
The last type of mitigation is crowd control. Melee sets with AoE knockdowns are popular for their extra mitigation provided. Unfortunately, this approach does not work on most Arch Villains, which can be considered the toughest challenge in tanking.
Some attack mechanics to consider:
A typical even level minion has a base of 50% chance to hit you. The minimum chance to hit is 5%. So, if you obtain 45% defense, we can conclude that you mitigate 90% of the damage you would have otherwise taken: 45 / 50 = 90
The resistance cap is also 90%. At first, it sounds like both forms of passive mitigation are equal. However, accuracy bonuses are given to critters (and players) for being higher level than the target and for ranks above minion. Accuracy is calculated after to-hit and defense, which means no amount of defense can make up for that bonus accuracy. Pure defense ends up being slightly less effective at mitigating damage than pure resistance. However, every defense set has some resistance and has access to Tough from the Fighting pool.
In the end, every set can do their job at tanking. It really comes down to team makeup, strategy, and player skill more than the set you choose.

Resist debuff has very little additional effect on Res-based characters- as mentioned, almost nothing has "unresistable" res debuff. Longbow grenades used to in the 40+ game, but I believe they changed that a year or two back. (I remember jumping into a pool of Longbow on an Invuln brute, shortly after they introduced real number/real time monitoring, and being quite surprised at seeing my 30%-ish Defense and 80%-ish Resists all go into the red at once. )

but overall you have much much more -def than -res...
really defense vs resist comes down to this
defense toons hope to avoid the hit and thus are less 'tough' overall
while resistance toons know they are going to be hit and are tougher for it

Off the top of my head, foes that hit you with -resists include:
Longbow Nullifiers. If you're a vill, you'll see a lot of these. They now exist hero-side in some tip missions. Each one in a spawn will fire the Disruption grenade, so this adds up fast and is quite dangerous. Fortunately, there's an easy countermeasure - move. These debuffs are just location based fields.
All Wailers. Fighting these guys taught me a valuable lesson about the effectiveness of -resist debuffs. You get hit by one of their Sonic attacks and you think "oh that sort of tickled, didn't do much to my hp though. After a couple more hit you say "whoa, how'd my health get into the yellow so fast"? Shortly after that you get to select whether to rez in the hospital or your base. Please note - I'm not saying these guys are difficult, I'm just saying they can take you from green to dead very quickly if they get to stack enough sonic attacks.
Goldbrickers. Same as Wailers really. Except bosses seem to have SS, so while minions and lts hit you with Sonic, the boss gets to pack a mean punch.
CoT Frost Demons have Sleet.
Seers can be added to the list. Surveillance sucks if you're not the one using it.
It is a pretty amazing disparity between blue and red side for this.

I have LOTS of toons, some Resist, some Def, some both like Invul and WP. I know that being soft capped on Def is the end-all be-all for Def (like Elude) and Granite is pretty much the pinnacle of Resist.
I also know that there's LOTS of -Def in the game. How about -Resist? I actually prefer Resist toons to Def ones for some reason but I wonder how much -Resist is out there and in what form?
