Quickest would depend on what's available, if you still have access to your old computer's hard drive, copying the game folder onto the new computer (either through network filesharing, burning on to some sort of removeable media (flash drive, dvd, etc), or connecting old computer's hard drive to new computer), download the NCsoft launcher, and when it asks you where you want to download/install CoH, point it at the copied folder.
Not 100% certain on the steps involved with the NC Launcher, only dealt with that process in the initial switchover from the old updater.

If you have access to the old machine (or hard drive), start by running the installing and running the launcher on the new machine, then click to install city of heroes. Once it starts, cancel it and copy over your entire city of heroes directory. Then fire up the launcher again.

I just did a fresh install of CoH using the launcher and it only took about 15 minutes total.
