Yes, they play. They are almost never on their "dev" accounts so you wouldn't know you were teamed with one even if you were. Some of the "higher-ups" have been known to play on Justice - however, they play on I'm guessing every server.
As far as "signature characters," those characters are represented in-game by a specific build. Like you mentioned, BaBs is an Invuln/SS Tank. Castle's a Fire/Fire Blaster, Ghost Widow is a Dark/Dark Corruptor (or a variation thereof), and so on.

Honestly If I were a Dev playing this game I'd welcome the anonymity. There are far too many people who want to give them grief in their official capacities. They don't need the hassle in their "free" time.
