A Brute or a Mastermind would be my suggestions for a quick levelling villain. Both solo very well, and both contribute to teams. Grouping is generally faster, but it depends on the group.
Raising the mission difficulty will increase the level of mobs in your missions, and/or the size of the mobs in your missions. Here's the catch: it's only 'better' xp if you can handle it. Raising it to difficulty 5 and getting killed left and right and taking forever to finish missions isn't going to get extra xp no matter how much each guy drops. My suggestion is to run at level 1 for awhile, and when that gets easy, try level 2. Levels 2 and 4 give bigger mob sizes, 3 and 5 raise the level of the mobs. My preferred level is 4: more enemies, but fewer bosses, lets me chew through minions for fast xp.
A brute should be able to handle the 4th difficulty level pretty early on. I suggest something like a Electric Melee/Willpower for ease of play and good AoE damage (AoEs go through minions faster than single target does)
If a brute doesn't suit your playstyle try a Bots/Dark Mastermind.

I made a Super Strength/Willpower brute recently and I have to say that it levels extremely fast. It also works really well as a solo character, has decent survivability that gets higher as you go up in levels, and can solo high difficulties without much issue.
Regarding difficulty settings, a lot depends on your power set choices. If you have a lot of area effect attacks then the second and fourth settings are great because they give more enemies per spawn. If you have high single target damage, the third and fifth settings are better because they give the same number of enemies, but at higher levels.
Personally, I'd go with a Brute if I was new. Masterminds are great, but they start out slower and are a bit trickier to play. With a Brute, you just run in and smash stuff... it's fast, fun, and very quick to level. Just be sure to slot for accuracy and endurance at low levels... you get most of your extra damage from Fury rather than enhancements, so not missing or running out of endurance is the important thing.
My suggestion is to go with a /Willpower Brute. They are good all-around characters, and Willpower is a "turn it on and forget it" defensive set that lets you concentrate entirely on smashing things. For a primary, you can't go wrong with Super Strength... it gives you good single target damage, some protection in the form of knockdown, and a fantastic AoE once you get to level 32. Dual Blades is also a lot of fun, though Dual Blades / Willpower is a very tight build since you'll want almost every power from both sets. The main thing is to pick something you find fun, because getting to 50 a little faster isn't worth not enjoying the trip.

I didn't love my Electric brute until level 33.
I have a Fire/Will that has been a lot of fun from pretty much level 1 to present (level 29, I think.)
I recommend an axe/will brute to a friend of mine and she loved, loved, loved it. That was what got her to 50.
Now that doesn't mean YOU will like it, but it was fun for me.
Note: All the villains I have tried seem to have a slow spot around level 16-21. You just have to fight through it.

I'd suggest trying to not rush to 50 for an AT you have no solid proof fits your playstyle. Find out what sort of Arachnos "spider" you want to play, find out what AT is somehwat close to that, then play that second AT - not for quick leveling, but to play for fun. Fun NOW, not later.
If you enjoyed it, then come 50 you might enjoy the unlockable AT. But starting a game to rush through it to then play it with something else seems a bit... pointless to me.
I'm new to the game and I have been trying to figure out what AT I want to be. As I was going through all the info on the forums I ended liking the spider AT's. Then I found out I have to have a 50 villian to even make one. If I'm looking to lvl to 50 as fast as possible what would be the best AT and should I solo or grp? I prefer to solo but would grp if it's slot faster. Also does raising mission difficulty make you lvl faster or does it slow you down more then give you extra exp?
