Use a character that can mow down large numbers of minions/LTs quickly and efficiently, get a mission full of mobs that are relatively easy to kill (Nemesis a good choice for villains, BM guys for heroes)
Then to up the spawn count you can do one of several things. You can either just run it solo on the second or fourth difficulty so you get larger spawns or you can get fillers each run.
Another alternative is getting a few friends to bring characters they don't plan on playing a lot into an Ouroboros arc that has your farm mission in it, then having them log off. The mission will still spawn for the teamsize, even if you're the only person online. -

And how does it work in say Health? Does it work like the p.Shifter proc, except it gives you a heal and endurance?

Honestly you guys I'm surprised no one has mentioned that a year ago it cost him over a billion inf to get a build with 3 purple sets and 7.5s/uniques.
That was my first clue that the OP fails at the market. -

Yea our team was overkill on the defense. In addition to the 3 Widows/Forts, we had a Cold and a Bubbler.
Also, since I was the one "tanking" on my Fort I made sure I had Frostworks kept on me and the strange thing is it showed my HP went up to 2103.1. I'm not sure if it was just a display bug since isn't the VEAT HP cap the same as Blasters?

Well uh thats kinda the thing. Widows/Forts often provide more defense than Crabs due to Mind Link. You don't need someone to tank alphas when everyone is at the defense soft-cap.

The cold shields don't benefit from Power Boost because they have a +resist portion (cold, fire). I'm not sure about Frostworks but I wouldn't have thought so. That's because its not a heal and a +hp like Dull Pain, its just +hp. In addition to that it has a +resist portion (toxic) so I doubt it would work anyway with PB.

Edit: not to downplay it, any MoRSF is a feat, just sayin' -
I would recommend swapping the Stupefys in Fissure out for Positron Blasts or Ragnaroks (without the damage proc). You'll get the same recharge bonus (or more with Rags) and Stupefy isn't giving any s/l defense so its not a big deal.
I would also swap out the generic Acc and Recharge IOs in Seismic Smash for the quad and triple from Mako's Bite, you'll get much better damage enhancement as well as higher values for acc end and rech.
Might also want to consider a PBAoE melee set in Hot Feet, you're not enhancing its damage much and 3% rech for 5 slots is an awful deal. Once you can afford it swap the Stupefys in Flashfire out for Absolute Amazements and you can drop the recharge IO for use elsewhere.
You also don't need two slots of resistance in Tough. Choosing to enhance its low resist value when you've got softcapped s/l defense already seems superfluous and those slots could be put to better use elsewhere. -
For me personally, going back to my defenders having played corruptors for a long time I barely noticed any increase in buff/debuff strength, but I did notice the severe damage drop.

While the disparity between +def and +res bonuses is huge and you raise a very good point for why this is the case Fulmens, it seems a little restrictive.
It would be cool if you could get +def -or- +res from set bonuses, so defense based characters could get more defense and resist based characters could shore up their resists. I'm not sure how this could be elegantly implemented though. The problem is I suppose heavily IOd builds could also get a decent amount of both which as you've said is overpowered. -
