There are three of them, at 24, 34 and 44. The trial's different for hero and villainside. You do require a team (of four, I believe.)

You can also get them (a) by buying/getting a drop of a Respec recipe, (b) getting some veteran respecs, or (c) the devs will *likely* give respecs out at issue 21's release.

And of course if you don't want to deal with any of that, at level 10 you can use your second build. (Once you unlock alpha, you can use a third.) It does need to be enhanced, etc. separately.

Difference.
There are three kinds of "Freespecs"
Devs hand one out.
The Respec Recipes (Both inventions and event vendor) are another kind.
And the bought-with-real-money ones as I recall.
A character can only have one freespec of any kind. On the plus side, teh crafted ones grey out if you have one.
