San Antonio, Texas
Off the top of my head, I'd suggest Sea World, the River Walk, and the Alamo. (Make sure you check out the basement!)
I've only been to San Antonio a few times, but those were the things I remembered most.

Miss Informed in 2016! She can't be worse than all those other guys!
I was stationed in San Antonio in the mid-80's. Great town.
The riverwalk is great. Make sure to visit Dirty Nellie's, if it's still there. If you have a car and a chance to see the countryside, the Hill Country up around Kerrville is lovely.

The food is farking fantastic. You will leave very full.
Be well, people of CoH.
Every time I hear of San Antonio, I can't help but think of this.
The Alamo (the actual building) is decidedly underwhelming. If you like history, it's a must, but don't expect an awe-inspiring structure. How else do you think Ozzy accidentally peed on it? Well, that and all the drugs.
Have fun, the city itself is awesome. The Riverwalk is probably worth a few visits. I'd consider making a day trip to Austin if I were going. Actually...can I come, too? I fit nicely in the trunk of most compacts.

Sorry I just now saw this.
If I were you I'd stay in New Braunfels and make day trips into San Antonio (32 miles to the Alamo/downtown) to see the sights there. Meanwhile you can float in the Comal and/or Guadalupe rivers and make a day of it at Schlitterbahn which is just about the most awesome water park in the world. The Natural Bridge Caverns is also nearby and worth a few hour's diversion.
The city is fairly small and built around tourism so there are plenty of places to stay - we usually rent a condo on the river when we make weekend trips down there.
In your trip to San Antonio I'd suggest you do the whole mission trail and not just the Alamo. The other four missions are in much better shape and very cool to see.
We are planning to visit San Antonio in summer 2010 and was wondering if anybody could give me advice/tips on what to do, where to stay, etc.
