Does the Bruiser need endurance reduction?
I still think so, but YMMV, especially if you choose to not give him one of the upgrades.

I plan on using both of the upgrades.
My planned slotting is: 3 Hami acc/dmg, Chance for BU, 10%Res, 5%def.
So my question boils down to, would his endurance be okay with this slotting?
If you are constantly fighting, yes he will have issues.
It also depends on your secondary. If you are /PD, painbringer will fix that problem. Also, if you routinely group with a /kin or /rad, their buffs can help. If solo and you chainpull then an End reduct is a good idea.
Bruiser and Enforcers both love some endredux slotting (and the Bruiser also loves the Explosive Strike damage proc since almost all his attacks do KB/KD of some sort).

3 Acc/Dmg Hamis
1 IO END Reduction
1 10% Res All
1 Build Up Soulbound

If you pick up Gang War there's no need to slot the 10% Resistance to all into the Bruiser actually. And 1 IO Endurance reduction won't exactly cover his hefty endurance costs sadly, but it will take quite the long fight(s) to smack down his blue bar.
My 2 cents.
I'd drop the resist bonus and slot that into something else like GW as Jay said. I'd also remove the soulbound considering the bruisers animations make the proc useless.
If you want bonus damage slot Explosive Strike: Chance for Smashing Damage. You will get a lot more out of the damage proc then the buildup proc IMO.
Mind you if you slotted like this, you'll be a lot happier with your Bruiser.

I moved my chance for build up into my Call Thugs, considering their are 3 of them. Also, I use both the 10% from recharge set and the one from pet damage, making 20% total.
42% end redux will not be enough for long fights (such as AV soloing).

5 Slots, won't break the bank. Never use a single stat IO if you can avoid it. 2 x Dam/Acc is better then 1 x Acc and 1 x Dam.
I know awhile back, when I played my 50 Mercs/ MM that the commando needed an endurance reduction in his slotting, which would lower the end cost of his moves he uses. My commando was consistantly at no endurance until I slotted 1.
Is the bruiser the same way? Or have they changed the slotting of endurance reductions?
